Spermidine

Also known as: Spermidine Trihydrochloride, SPD, Polyamine Spermidine

Spermidine is a naturally occurring polyamine found in all living cells, with exceptionally high concentrations in wheat germ, aged cheese, and human sperm. It is the most studied autophagy-inducing dietary compound, shown to extend lifespan across multiple species and reduce cardiovascular and cognitive aging.

Half-Life

~30–60 minutes, but gut bacteria produce it continuously; supplementation raises tissue levels over weeks

Key Benefits

  • Induces autophagy — cellular self-cleaning
  • Extends lifespan in yeast, flies, worms, and mice
  • Reduces cardiovascular aging and arterial stiffness
  • Reduces all-cause mortality (human epidemiological data)
  • Neuroprotective: reduces amyloid and tau pathology
  • Promotes hair growth (anagen phase activation)
  • Reduces age-related immune decline
  • Improves memory in aging models

Mechanism of Action

Spermidine induces autophagy — the cellular 'self-cleaning' process that removes damaged proteins, organelles, and intracellular debris. It inhibits EP300 acetyltransferase, leading to hypoacetylation of histones and activation of autophagy genes (ATG5, ATG7, Beclin-1). In the cardiovascular system, spermidine reduces arterial stiffness by preventing cross-linking of extracellular matrix proteins. In the brain, it promotes mitophagy (removal of dysfunctional mitochondria), reduces amyloid-beta accumulation, and maintains synaptic plasticity. Spermidine levels decline with age, correlating with reduced autophagy and accelerated aging.

Side Effects

  • Generally very well tolerated
  • Rare: mild GI discomfort at high doses
  • May temporarily reduce some gut bacteria species
  • Rare: headache at initiation

Contraindications

No significant contraindications. Wheat germ-sourced supplements: avoid in celiac disease unless certified gluten-free. Insufficient data in pregnancy.

Storage
