Pal-GHK

Also known as: Palmitoyl GHK, Palmitoyl Tripeptide-1, Pal-GHK (non-copper)

Pal-GHK is the palmitoylated form of the GHK tripeptide without a copper ion. By conjugating palmitic acid to glycine-histidine-lysine, skin penetration is substantially enhanced, enabling deeper dermal collagen stimulation. It is commonly paired with Pal-GHK-Cu or GHK-Cu in anti-aging formulations.

Key Benefits

  • Stimulates collagen I and III synthesis in dermis
  • Reduces the appearance of fine lines and wrinkles
  • Improves skin elasticity and firmness
  • Inhibits collagenase (MMP-1) to preserve existing collagen
  • Enhances wound healing and skin repair
  • Well-tolerated in anti-aging serums and creams

Mechanism of Action

Pal-GHK enhances dermal penetration via the palmitate lipid tail and signals fibroblasts to upregulate collagen I, III, elastin, and glycosaminoglycan synthesis. It activates TGF-β and insulin growth factor receptor pathways, increases fibronectin production, and inhibits MMP-1 (collagenase), reducing collagen degradation. Without copper, the mechanism is growth factor signaling-dominant rather than metalloenzyme-mediated.

Dosing Protocols

Anti-Aging (Standard)

Dose
0.005–0.1% in formulation
Frequency
Once or twice daily
Timing
PM primarily; AM with SPF acceptable
Cycle
12–24 weeks, ongoing

Often formulated alongside Pal-GHK-Cu and Matrixyl (Pal-KTTKS) for a comprehensive anti-aging peptide complex. Avoid strong acid exfoliants in the same step to preserve peptide integrity.

Side Effects

  • Generally very well-tolerated
  • Rare skin irritation at very high concentrations
  • Possible formulation-dependent comedogenicity

Contraindications

Known sensitivity to palmitoyl esters or GHK peptideActive skin infections

Storage

Store in a cool, dark location. Sensitive to oxidation and hydrolysis; sealed airless pump containers preferred. Refrigerate after opening for extended shelf life.
