Nonapeptide-1

Also known as: Melanostatine-5, MFML peptide, melanogenesis inhibitor peptide

Nonapeptide-1 is a synthetic 9-amino acid peptide that inhibits melanin production by blocking α-MSH (alpha-melanocyte stimulating hormone) receptor binding. Used in cosmetic formulations for skin lightening and evening skin tone, it is particularly effective for UV-induced and hormonal hyperpigmentation.

Key Benefits

  • Inhibits UV-induced tanning and hyperpigmentation
  • Reduces hormonal melasma
  • Evens skin tone at receptor level
  • Well-tolerated with minimal irritation
  • Complementary to tyrosinase inhibitors for enhanced brightening
  • Reduces post-inflammatory hyperpigmentation

Mechanism of Action

Nonapeptide-1 is a competitive antagonist of the MC1R (melanocortin-1 receptor) on melanocytes. By blocking α-MSH binding, it prevents cAMP elevation and the downstream PKA activation that normally drives MITF expression and tyrosinase synthesis. This receptor-level blockade reduces melanogenesis more upstream than tyrosinase inhibitors like Decapeptide-12, providing complementary depigmentation activity.

Dosing Protocols

Skin Brightening (Standard)

Dose
0.05–0.5% concentration in formulation
Frequency
Twice daily
Timing
AM and PM after cleansing
Cycle
8–16 weeks

Combine with SPF 30+ during daytime use. Can be stacked with Decapeptide-12 or vitamin C for multi-pathway brightening. Consistent daily use required for visible results.

Side Effects

  • Generally very well-tolerated
  • Rare contact sensitivity in susceptible individuals
  • Theoretical risk of excessive depigmentation with prolonged high-concentration use

Contraindications

Known peptide allergyCaution in pregnancy (limited safety data)

Storage

Store at room temperature, away from direct sunlight and heat. Stable in pH 4–7 formulations. Avoid exposure to air oxidation; prefer airtight packaging.
