Decapeptide-12

Also known as: Luminixyl, 10-amino acid depigmentation peptide

Decapeptide-12 is a synthetic 10-amino acid peptide developed for skin brightening and depigmentation. It selectively inhibits tyrosinase activity and downstream melanogenesis pathways, reducing hyperpigmentation, dark spots, and uneven skin tone without the irritation associated with hydroquinone.

Key Benefits

  • Reduces hyperpigmentation and dark spots
  • Evens skin tone and improves radiance
  • Inhibits post-inflammatory hyperpigmentation
  • Well-tolerated alternative to hydroquinone
  • Effective for melasma and age spots
  • Non-cytotoxic to melanocytes

Mechanism of Action

Decapeptide-12 competitively inhibits tyrosinase, the rate-limiting enzyme in melanin biosynthesis, at the copper-binding active site. It also downregulates MITF (microphthalmia-associated transcription factor) expression and reduces tyrosinase-related protein-1 (TRP-1) and TRP-2 levels, providing multi-pathway inhibition of melanogenesis. Unlike hydroquinone, it does not cause melanocyte cytotoxicity.

Dosing Protocols

Skin Brightening (Standard)

Dose
5 ppm (0.0005%) concentration
Frequency
Twice daily (AM and PM)
Timing
Apply after cleansing, before moisturizer
Cycle
12–16 weeks minimum

Clinical studies used 5 ppm; often found at 0.001–0.01% in commercial serums. Pair with SPF during daytime use to prevent UV-induced repigmentation. Can be layered with vitamin C for enhanced brightening.
