Argireline

Also known as: Acetyl Hexapeptide-3, Acetyl Hexapeptide-8, AH3

Argireline is the most widely researched topical 'Botox-alternative' cosmetic peptide, an acetylated hexapeptide that inhibits neuromuscular transmission to relax facial muscles and reduce dynamic wrinkle depth by 17–27% in clinical studies.

Key Benefits

  • Reduces depth of dynamic expression wrinkles 17–27%
  • Non-invasive topical Botox alternative
  • Smooths forehead, eye area, nasolabial fold lines
  • Widely studied — published clinical efficacy data
  • Synergistic with SNAP-8 for enhanced effect
  • Reduces fine lines around eyes (crow's feet)
  • Improves skin smoothness and texture
  • Well tolerated across all skin types

Mechanism of Action

Argireline (Acetyl-Glu-Glu-Met-Gln-Arg-Arg-NH2) is a fragment of SNAP-25, a SNARE complex protein required for neurotransmitter vesicle docking. Argireline competitively inhibits formation of the SNARE complex at neuromuscular junctions, reducing acetylcholine release and thus muscle contraction intensity. The result is partial relaxation of facial expression muscles without paralysis — a mechanism similar to Botox but topically delivered at lower efficacy. Optimal concentrations are 5–10%; higher concentrations may cause temporary ptosis.

Dosing Protocols

Anti-Wrinkle Serum

Dose
5–10% concentration
Frequency
Twice daily
Timing
AM after cleansing / PM before bed
Cycle
Ongoing; visible results in 28–56 days

Avoid >10% — can cause temporary muscle droop. Combine with SNAP-8 at 3–5% for synergy

Eye Cream / Crow's Feet

Dose
5–8% concentration
Frequency
1–2x daily
Timing
Around eye area, tap gently
Cycle
Ongoing

Allow to dry before applying moisturizer on top

Side Effects

  • Generally very well tolerated
  • At >10%: temporary eyelid/brow ptosis (drooping)
  • Rare: mild redness in sensitive skin
  • No systemic absorption at cosmetic doses

Contraindications

External use only. Not for injection. Keep below 10% concentration. Avoid eye contact. Not studied in pregnancy (no known risk but insufficient data).

Storage

Active ingredient: 2–8°C. Formulated products: cool, dry place per product label. Typically stable for 24 months.
