Alpha-GPC

Also known as: Alpha-glycerophosphocholine, L-Alpha glycerylphosphorylcholine, Choline Alfoscerate

Alpha-GPC is the most bioavailable form of choline, readily crossing the blood-brain barrier to rapidly increase acetylcholine levels. It enhances cognitive performance, supports GH secretion, and is used as an essential complement to many nootropic peptides (especially those that increase acetylcholine demand like Noopept and Dihexa).

Key Benefits

  • Rapidly raises brain acetylcholine levels
  • Enhances memory formation and recall
  • Prevents headaches from nootropic peptides (choline donor)
  • Stimulates growth hormone secretion (modest)
  • Improves attention and processing speed
  • Neuroprotective in Alzheimer's and cognitive decline
  • Approved in Europe for Alzheimer's therapy
  • Enhances power output in athletes (pre-workout)

Mechanism of Action

Alpha-GPC is hydrolyzed in the gut to glycerophosphate and choline. The choline fraction is efficiently transported across the blood-brain barrier and incorporated into neuronal membranes as phosphatidylcholine, and used as the precursor for acetylcholine synthesis via choline acetyltransferase (ChAT). Unlike other choline supplements (choline bitartrate, CDP-choline), Alpha-GPC has higher bioavailability (biological availability ~88% vs 1-3% for choline bitartrate). In older subjects and Alzheimer's models, Alpha-GPC improves cholinergic transmission and has been shown to stimulate growth hormone secretion via HPA axis modulation.

Dosing Protocols

Nootropic Choline Support

Dose
300–600 mg
Frequency
1–2x daily
Timing
Morning and/or before cognitive tasks
Cycle
Ongoing

Essential pairing with Noopept, Dihexa, and other cholinergic nootropics to prevent depletion headaches

Pre-Workout / GH Protocol

Dose
600 mg
Frequency
Once daily
Timing
30–60 minutes pre-workout
Cycle
Ongoing

At 600 mg, studies show improved power output; pairs well with GH-releasing peptides

Side Effects

  • Headache (paradoxically, from excess acetylcholine at very high doses)
  • Nausea at doses > 1200 mg
  • Dizziness
  • Fatigue at high doses
  • Rare: depression with chronic very high dosing

Contraindications

Avoid in cholinergic hypersensitivity. Caution with scopolamine or anticholinergic drugs (may reduce their effect). Avoid in known sensitivity to choline or lecithin.

Storage

Store at room temperature in airtight container. Alpha-GPC is hygroscopic (absorbs moisture) — keep sealed. Capsules preferred over powder for stability. Typically 2-year shelf life.
