AICAR

Also known as: 5-Aminoimidazole-4-carboxamide ribonucleotide, Acadesine, AICA riboside

AICAR is a cell-permeable AMP analog that activates AMPK (AMP-activated protein kinase) — the master metabolic switch that triggers fat burning, mitochondrial biogenesis, and adaptations normally only achieved through exercise. It has been called the 'exercise in a pill' compound.

Key Benefits

  • AMPK activation mimics aerobic exercise adaptations
  • Increased fat oxidation and endurance
  • Mitochondrial biogenesis (PGC-1alpha)
  • Improved insulin sensitivity and glucose metabolism
  • Anti-inflammatory effects
  • Potential cardiac protection during ischemia
  • Synergistic with actual exercise training
  • Reduces hepatic glucose production

Mechanism of Action

AICAR is converted intracellularly to AICAR monophosphate (ZMP), a potent AMPK activator. AMPK activation triggers a cascade mimicking exercise adaptations: increased fatty acid oxidation, enhanced glucose uptake in muscle, inhibition of fat synthesis, mitochondrial biogenesis via PGC-1alpha activation, and improved insulin sensitivity. In animal studies, AICAR administration increased running endurance by 44% without exercise training, making it WADA-prohibited.

Dosing Protocols

Research Protocol

Dose
25–50 mg
Frequency
3–5 times per week
Timing
Any time, subcutaneous
Cycle
4–8 weeks

WADA prohibited substance — banned for competitive athletes. Limited human dosing data. Animal studies used 0.5–1 mg/kg doses. Human translational doses are estimated. Stacking with exercise amplifies metabolic benefits.
